【摘要】 采用小鼠S180移植瘤實驗研究了巴西菇子實體中麥角甾醇的體內(nèi)抗腫瘤活性,并通過巨噬細胞吞噬功能測定(中性紅法)、脾淋巴細胞增殖功能檢測(MTT法)、體外抑殺腫瘤細胞(MTT法)以及雞胚絨毛尿囊膜(CAM)等試驗初步探討了麥角甾醇抗腫瘤活性的作用機理。結(jié)果表明,巴西菇子實體中提取的麥角甾醇在劑量10 mg.kg-1.d-1、50mg.kg-1.d-1和100 mg.kg-1.d-1時的瘤重抑制率分別為62.75%、76.61%和79.18%,說明麥角甾醇有較強的抑瘤活性。在巨噬細胞吞噬試驗和脾淋巴細胞增殖試驗中,麥角甾醇各劑量組與陰性對照組相比無統(tǒng)計學(xué)差異(p>0.05),沒有劑量依賴趨勢,說明其對巨噬細胞吞噬功能和脾淋巴細胞增殖均無明顯促進作用,可能不是通過增強免疫來發(fā)揮抑瘤活性的;麥角甾醇體外對人胃癌細胞SGC-7901和肝癌細胞Bel-7402的抑制試驗中,與對照組相比均無統(tǒng)計學(xué)差異(p>0.05),說明其抑瘤活性也不是通過細胞毒作用來發(fā)揮的;在CAM實驗中,麥角甾醇與相同劑量(5μg.mL-1)的陽性對照地塞米松相比,表現(xiàn)出了更強的抑制血管生長的作用(抑制率63.46...
